台灣地區觀賞魚疾病研究

台灣是世界上觀賞魚飼養的重要地區,台灣的位置位於熱帶亞熱帶,常年可保持熱帶魚生長所適合的氣候,天然環境及水質的條件適合多種觀賞魚蝦的繁養殖,且傳統中華文化飼養觀賞魚具有悠久的歷史及長時間繁養殖技術的累積,使得台灣觀賞魚市場蓬勃發展,政府並在屏東農業生物科技園區設置包含自研發、蓄養、檢疫、包裝、乃至進出口報關作業的「亞太水族營運中心」,期望能簡化並加速觀賞魚進出口流程,協助外銷拓展。本次研究希望調查台灣民眾飼養不同觀賞魚時較常發生或遭遇到的疾病及狀況,自台灣各地區魚場、個人養殖戶及進口中盤商收集樣本,分類為鱸形目攀鱸亞目、鱸形目鱸亞目、鱸形目隆頭魚亞目、鱸形目雀鯛科、鯰形目、脂鯉目、鯉形目、虹

銀漢魚目、骨舌魚目、鱂形目、燕魟目、魨形目及觀賞蝦。自104年8月至106年4月共收集237個觀賞魚病例,魚隻品種共74種,在232個病例中有104例(34%)檢出細菌、86例(29%)檢出寄生蟲、59例(20%)檢出病毒及52例(18%)包含水質不良等其他原因。細菌檢出前三高者為Aeromonas sobria 26例(24%)、Mycobacterium spp. 21例(20%)、Vibrio harveyi 11例(10%)。寄生蟲檢出前三高者為吸蟲類的33例(38%)、白點蟲13例(12%)、車輪蟲及四膜原蟲各為10例(10%)。以Lamp方式檢測錦鯉皰疹病毒(KHV)有21例(41

%)呈現陽性、以PCR方式檢測則有1例(2%)呈現陽性。以PCR方式檢測虹彩病毒(含GIV、TGIV、RSIV、ISKNV)有40例(62%)呈現陽性。依照本研究統計,觀賞魚最好發疾病為細菌性病原引起,淡水魚為Aeromonas sobria,海水魚則為Mycobacterium spp.,針對淡水魚產氣單胞菌屬細菌造成觀賞魚大量疾病,本研究同時對收集到的菌株進行藥物敏感性試驗。感染病原其次為寄生蟲性疾病,體外寄生的吸蟲類為最多,如指環蟲及三代蟲。台灣目前仍為錦鯉疱疹病毒之疫區,根據本研究統計多數為潛伏感染,虹彩病毒雖有超過半數的檢出,但其對觀賞魚的影響則尚須研究探討。

人類諾羅病毒是急性腸胃炎重要致病原之一,已在全球造成流行,並且台灣自2002年以來陸續有病例報導。目前病毒診斷技術有靜態描述定性法與動態變化定量法並已由定性法進展到定量法。本論文研究目的如下:一、利用酵素結合免疫吸附法和逆轉錄聚合酶鏈鎖反應法檢測諾羅病毒之方法比較與評估。二、利用逆轉錄聚合酶鏈鎖反應(定性)法來探討2011-2012年台灣地區諾羅病毒感染的盛行率與病毒基因型別。三、利用即時定量逆轉錄聚合酶鏈鎖反應(定量)法來探討2012-2013年台灣地區諾羅病毒感染的病毒量與疾病病徵嚴重程度之相關性。 首先,本研究使用已上市之第三代酵素結合免疫吸附法及逆轉錄聚合酶鏈鎖反應法針對急性腸胃炎

及非急性腸胃炎患者糞便檢測諾羅病毒。本研究自2011年8月起至2012年7月,共收集253個糞便樣本,利用上述兩項方法學予以檢測分析及評估比較。本研究結果證實,當酵素結合免疫吸附法與逆轉錄聚合酶鏈鎖反應法比較時,在全體樣本、急性腸胃炎樣本、非急性腸胃炎樣本、小於10歲及大於10歲之檢測靈敏度,分別為20.8%,23.5%,14.3%,40%及7.1%;特異性分別為99.6%、99.3%、100%、100%及99.1%。本研究亦證實,利用酵素結合免疫吸附法檢測分析全部糞便樣本,有6個陽性樣本(陽性率2.4%);使用逆轉錄聚合酶鏈鎖反應,有24個陽性樣本(陽性率9.5%)。本研究證實,第三代酵素結

合免疫吸附法比逆轉錄聚合酶鏈鎖反應法,更具低靈敏度與高特異性,並且這兩種方法檢測諾羅病毒之陽性率亦不相同。 接著本研究即利用逆轉錄聚合酶鏈鎖反應法針對感染諾羅病毒患者之分子流行病學及親緣分析。本研究自2011年8月至2012年7月,共收集253患者糞便樣本(包括155個有症狀和98個無症狀),利用逆轉錄聚合酶鏈鎖反應法檢測諾羅病毒陽性與否。再以逆轉錄聚合酶鏈鎖反應產物經DNA定序後作親緣分析。結果顯示利用逆轉錄聚合酶鏈鎖反應法檢測 253個糞便檢體,共有24個(9.5%)諾羅病毒陽性個案。諾羅病毒株分離自所有年齡層(1至86歲)並且分離出諾羅病毒陽性樣本79.2% (19/24)是來自於住院
