雜環胺(heterocyclic amines, HAs)是由碳、氫及氮原子所組成,具有多環芳香結構的化合物。QuEChERS為一種快速樣品前處理技術,其效果會受淨化粉劑組成所影響。本研究目的為發展超高效液相層析配合串聯式質譜儀分離肉品中21種HAs之方法,及探討不同QuEChERS淨化粉劑組合對萃取肉品HAs之效果。結果顯示,YMC-Triart C18 ExRS管柱(150 mm × 2.1 mm i.d., 1.9 μm),動相A為濃度10 mM的醋酸銨緩衝溶液(pH 4.75)、動相B為氰甲烷,搭配梯度條件0-0.4分鐘(95% A),0.4-2.3分鐘(60% A),2.3-2.5

分鐘(20% A),2.5-6.5分鐘(20% A),6.5-6.6分鐘(95% A),流速為0.4 mL/min且管柱溫度維持在35oC,配合三段四極體式串聯質譜儀(triple quadrupole tandem mass)以選擇反應監測掃描模式(selective reaction monitoring)偵測,可以在4.5分鐘內分析21種HAs,其中包含一種內部標準品(4,7,8-TriMeIQx),較其他管柱分離度好並具有短分離時間。於水煮、烘烤及油炸加工方式下之不同肉品中添加10 ng/g之20種HAs,再分別以含0、75及150 mg C18EC之QuEChERS淨化粉劑進行萃取,

發現以75 mg C18EC組別萃取可獲得水煮雞肉、烘烤雞肉、豬肉樣品及牛肉樣品中20種HAs之最高回收率分別為62.26-102.85%、60.58-127.54%、54.52-103.74%及63.83-110.20%;以150 mg C18EC組別萃取可獲得油炸雞肉和魚肉樣品中20種HAs之最高回收率分別為72.08-111.84%和56.51-103.76%。以上方法於75 mg C18EC組別的水煮豬肉萃取液之偵測極限及定量極限分別為0.001-0.05 ng/g及0.002-0.1 ng/g,日內精密度之變異係數(coefficient of variation, CV%)介於2.

揮發性硫醇 4-mercapto-4-methylpentan-2-one (4MMP) 是葡萄酒發酵過程中酵母菌的代謝產物,帶有黃楊木、黑醋栗般的香氣,感官閾值 (Threshold) 極低,為葡萄酒中獨特且重要的香氣化合物。然而市面上香料多以化學合成法製成,現今全球市場對於天然香料的需求逐年增加,故本研究評估不同酵母菌來源的 β-lyase 從半胱胺酸共軛前驅物釋放硫醇化合物的能力,預期以酵母菌細胞抽出物轉化的方式提高 4MMP 之轉換率,製備葡萄酒香氣化合物 4MMP。以線上蛋白質資料庫 (UniProt) 及蛋白質序列比對工具 (Basic Local Alignment Searc

h Tool, BLAST) 進行 BLAST 分析尋找具有 β-lyase 相似序列之同源蛋白,篩選出 Saccharomyces cerevisiae (3 株), Kluyveromyces marxianus, K. lactis, Debaryomyces hansenii, Schizosaccharomyces pombe 等七株酵母菌用於製備細胞抽出物。將人工合成之半胱胺酸共軛前驅物 cys-4-mercapto-4-methylpentan-2-one (Cys-4MMP) 以 Electrospray Ionization Mass Spectrometry (ESI-MS
